NLRP6 is a key innate immune sensor whose functions have remained understudied. In a recent Cell paper, Shen et al. evaluate the mechanism of NLRP6 activation and find that double-stranded RNA interacts with NLRP6 and leads to liquid–liquid phase separation that is critical for NLRP6-dependent innate immune signaling.
