Clonal hematopoiesis of indeterminate potential (CHIP) is a frequent health-debilitating condition resulting from progressive, age-dependent expansion of mutant hematopoietic stem and progenitor cells. In a recent Nature paper, Agarwal et al. show that aging associates with increased gut permeability and consequent higher circulating levels of a microbiota-derived metabolite, ADP-heptose, which upon engaging its receptor ALPK1, favors the clonal expansion of CHIP-mutant hematopoietic stem cells.
